Genisphere’s newest RNA Amplification kits provide a two-round protocol that is as easy and accurate as one round. In this process, Sense-Strand Amplified RNA is produced through a proprietary Tandem RNA Polymerase Promoter technology referred to as TRPP.

TRPP technology works by using a tandem RNA polymerase promoter oligo that includes T7 and T3 RNA polymerase promoter sequences. As a result, the amplification product from round one T7 In Vitro Transcription has a built-in T3 RNA polymerase promoter sequence which is utilized in round two to avoid repeating the entire “promoter synthesis” process. The round two double stranded T3 RNA polymerase promoter is simply generated in two steps. The template strand is synthesized during the second round reverse transcription reaction and the non-template strand is hybridized to the cDNA template as a short oligonucleotide.

Very similar to our SenseAMP product, RampUP also results in no 3’bias. Amplification begins by synthesizing cDNA from the 3’ end of the message. The round one T7 amplification reaction that follows initiates at 3’ end of the cDNA (originally the 5’end of the RNA).

RampUP Plus is recommended for laboratories using senseRNA in downstream dT priming applications. The “Plus” portion of RampUP Plus is for poly (A) tailing the senseRNA to ensure compatibility with any dT-based reverse transcription protocols.
